HCTU (CAS: 330645-87-9) is a novel aminium-based coupling reagent used during the process of solid phase peptide synthesis (SPPS), analogous to HBTU, which in comparative studies was found to give superiort results to TBTU in the synthesis of difficult peptides, in hindered couplings, and cyclizations. Peptide synthesis is important within chemistry, allowing us to create new peptides and bonds (this is the backbone of proteins).
HCTU is a highly efficient coupling reagent that could be used as a low-cost alternative to more expensive, highly efficient reagents such as HATU when fast coupling times (5 min or less) are desired. Fast coupling times promote increased productivity, and in some cases may also result in purer crude peptides by not allowing kinetically slower side reactions to compete.
HCTU is a coupling reagent used for amide and ester formation and commonly employed in peptide synthesis. HCTU is analogous to HBTU and has a Cl substituent on the phenyl ring in the 6 position. HCTU is non-toxic, stable in DMF, suitable for solid-phase and in solution synthesis. High purities, short reaction times and high coupling efficiency can be achieved when HCTU is employed, with performances comparable to HATU but at a more affordable price.
HCTU is a reagent for the synthesis of near-infrared pH activatable fluorescent probes and of human β-amyloid by Fmoc chemistry. HCTU is also a reagent for stereoselective Horner-Wadsworth-Emmons olefination, covalent ligation of fluorescent peptides to quantum dots, and alkylation of human telomere sequence.
